Peptide Sequence Identification and Confirmation by Advanced Analytical Techniques

Shlomy Arava, Analytical R&D, TAPI, 12, Israel (shlomy.arava@tapi.com)

Peptide sequence identification is essential for ensuring product integrity and consistency in peptide drug development. This presentation outlines an integrated analytical approach combining Amino Acid Analysis (AAA), peptide mapping, high-resolution LC-MS, LC-MS-MS, and NMR spectroscopy for comprehensive sequence confirmation. Each technique provides complementary structural information, from amino acid composition to sequence order verification. Case examples will demonstrate how orthogonal analytical data establish confidence in sequence identity and detect potential sequence variants or modifications. The presentation highlights best practices and method selection considerations for robust peptide sequence characterization.
